چکیده
Abstract Study question Is the redox state in embryo culture medium, as measured by oxidized albumin, associated with successful development and pregnancy outcome? Summary answer The level of albumin medium was most important variable predicting blastocyst formation andwas miscarriage after transfer. What is known already Human serum (HSA), abundant protein plasma, exerts antioxidant activities against oxidative damage. HSA exists human non-mercaptalbumin (HNA) reduced mercaptalbumin. HNA attracting attention because its novel role a marker reflecting state, levels are to increase stress diseases such kidney disease, diabetes, liver disease or Parkinson's disease. On other hand, IVF, supplemented source, but no previous studies have examined relationship between IVF outcomes. design, size, duration We followed outcomes for 40 cycles single transfer out retrospective study, which enrolled total 173 embryos cultured stage from 91 patients who underwent IVF/ICSI cycle February 2018 July 2018. Participants/materials, setting, methods Prior using culture, assessed level. level, patient age, oocyte maturity were analyzed factors on day 5 6 fertilization, machine learning model developed Random Forest (RF) algorithm predict formation. also whether made difference Main results chance median %HNA 89.36% (range, 80.12% 94.84%), much higher than value blood healthy humans, (approximately 25%). Blastocyst observed 41.04% (71/173) embryos. In both univariate multivariate analyses, lower (p = 0.001), younger age < use standard 0.007). A prediction RF four (%HNA, fertilizationmethod, maturation stage). 70% samples (training set, n 121) validated remaining testing set (n 52) produced an area under curve 0.761, where formation, age. significantly group that resulted (93.15% vs. 89.28%, P 0.0498). Limitations, reasons caution This study conducted G-TL only. Further needed elucidate association development, success rate commonly used media. Wider implications findings supposed mimic body environment, showed high compared itself; it suggesting may cause unsuccessful live birth rate. Controlling oxidation help create more appropriate environment. Trial registration number not applicable

O-8: Critical Role of Hyaluronan System in Pre-Implantation Embryo Development and Establishment of Pregnancy
Background: Hyaluronan (HA) is a structural component of extracellular matrix synthesised by HA synthases HAS1-3, which produce HA of different molecular sizes with distinct biological functions associated with reproductive processes. Hyaluronidase (HYAL) cleaves the HA into biologically active small fragments which are known to regulate cell proliferation through CD44 receptor signaling. HA is...
متن کاملIn vitro bovine embryo production in a synthetic medium: embryo development, cryosurvival, and establishment of pregnancy.
The aim of this study was to develop an in vitro embryo culture medium without either fetal calf serum or BSA, using various growth factors and cytokines (GFs-CYKs; IGF-I, IGF-II, bFGF, LIF, GM-CSF, TGF-β1, and PDGF-BB), and other molecules with surfactant and embryotrophic properties, such as recombinant albumin (RA) and hyaluronan (HA). The first part of the study was dedicated to define the ...
متن کاملThe Comparison Between the Pregnancy Outcome after Easy Embryo Transfer and Difficult Embryo Transfer
Background: The role of embryo transfer (ET) in the success of in vitro fertilization/intra cytoplasmic sperm injection treatment is not well understood. In the present study we classified ET technique as difficult and easy types according to certain strict criteria, and compared the pregnancy outcome of the two ET types. Methods: This study was performed retrospectively on 295 infertile patien...
متن کاملMitochondrial DNA content in embryo culture medium is significantly associated with human embryo fragmentation.
STUDY QUESTION Is the amount of cell-free DNA released by human embryos into culture medium correlated with embryo morphological features? SUMMARY ANSWER The mitochondrial DNA (mtDNA) content of culture medium is significantly associated with the fragmentation rate on Days 2 and 3 of embryo development, whether the oocyte came from women ≤ 35 or >35 years old. WHAT IS KNOWN ALREADY Cellular...
متن کاملP-127: The Effect of Supplementation of In Vitro Culture Medium with FGF2 on Ovine Embryo Development
Background: Fibroblast growth factors are a great family of growth factors that exert variety of functions in vertebrates including cell growth, survival, differentiation and migration. FGFRs are transmembrane tyrosine kinase receptors transmitting FGF signals. Four types of FGFRs (1/2/3/4) exist in vertebrates. FGF2 is one of the FGFs detected in bovine and ovine uterus flushes, supposing that...
